NanoDrop-style micro-volume spectrophotometers measure 1–2 µL drops; cuvettes measure 20 µL to 3.5 mL. Compare the two approaches and find the middle ground.
NanoDrop-style instruments (and competitors such as DeNovix DS-11, Implen, Thermo NanoDrop One) measure a 1–2 µL drop suspended between two pedestals, with a very short adjustable path length (0.1–1 mm). That lets them measure very high concentrations without dilution — ideal for DNA/RNA quantification from precious samples.
| Factor | NanoDrop (micro-volume) | Cuvette-based UV-Vis |
|---|---|---|
| Sample volume | 1–2 µL (drop) | 20 µL – 3.5 mL |
| Path length | 0.1–1 mm (adjustable) | Fixed, typically 10 mm |
| Dynamic range | Very high (up to ~1000× dilution range) | Standard (1–2 AU); dilute concentrated samples |
| Sample recovery | Yes (drop is retrievable) | Yes (fill and empty) |
| Throughput | Single drops, sequential | Batch with autosampler / multi-cell |
| Typical cost (instrument) | $8,000 – $15,000+ | $3,000 – $8,000 (entry UV-Vis) |
| Per-measurement cost | Low (no consumable) | Low (cuvette reusable) |
| Flexibility | Micro-volume assays only | All path lengths, fluorescence, kinetics, flow |
If you often have 50–200 µL of sample — not 1–2 µL, but not 3.5 mL either — a sub-micro cuvette gives you most of the volume saving without buying a second instrument. scuvette ultra-micro cells (20–200 µL, 10 mm path, JGS1 quartz) work in your existing spectrophotometer or fluorometer:
Many labs run both: a micro-volume instrument for quick DNA/RNA checks, and a cuvette spectrophotometer for everything else. If you can only afford one, a cuvette instrument plus 20–100 µL sub-micro cells covers ~90% of routine work — including the precious-sample cases.
