蛍光キュベット選択ガイド

Fluorescence is measured at right angles, which changes the キュベット you need. Here is 選び方 a セル that maximises signal and minimises background.

Fluorescence 分光法 detects emitted light at a right angle to the excitation beam. That one geometric fact drives almost every choice you will make about the キュベット — 窓 count, masking, 材質 and sealing.

1. Why Fluorescence Needs a 4-窓 Cell

A 標準 absorbance セル only needs two polished, opposing 窓s — the light travels straight through. Fluorescence, however, collects light perpendicular to the excitation 光路, so the side faces must also be optically polished:

  • 4-窓 セル — all four sides polished. Use for fluorescence, and also lets the same セル do absorbance along two axes.
  • 2-窓 セル — two polished faces only; fine for absorbance, but not for fluorescence.
  • 5-窓 セル — adds a polished bottom for bottom-reading or multi-angle instruments.

2. 材質 & グレード: JGS1 溶融石英

Fluorescence often targets UV-excited fluorophores, so the セル must transmit deep into the UV. JGS1 far-UV グレード 溶融シリカ transmits from ~190 nm through the visible range and is the 標準 for fluorescence work. Budget optical glass cuts off below ~320 nm and will block the UV excitation you rely on.

3. Black-Wall Cells for Low-容積 Fluorescence

Stray excitation light reaching the detector is the enemy of a clean fluorescence signal. A black-wall セル masks two opposite side walls (leaving the optical 窓s clear), which:

  • Blocks stray light from entering the detection 光路.
  • Lowers background and improves the signal-to-noise ratio.
  • Is especially valuable for low-容積 or low-concentration samples.

4. 光路長 & Sample 容積

  • 10 mm 光路 is the 標準 for fluorescence; it fits most fluorometers.
  • Semi-micro and sub-micro formats keep a 10 mm 光路 while reducing the sample 容積 to a few hundred microlitres — 最適 proteins and nucleic acids.
  • Confirm both the 光路長 and the external dimensions (12.5 × 12.5 mm is 標準) against your instrument holder.

5. Sealing: 標準 vs Screw-Cap

  • 標準 cap — fine for routine aqueous fluorescence measurements.
  • Screw cap (PTFE) — seals tightly for volatile, oxygen-sensitive or hazardous samples, and prevents evaporation during long kinetic runs.

6. Quick Checklist

  1. Fluorescence measurement? → 4-窓 セル.
  2. UV excitation below 320 nm? → JGS1 溶融石英.
  3. Low signal or tiny 容積s? → black-wall micro セル.
  4. Volatile or oxygen-sensitive sample? → screw cap.
  5. Check holder fit (12.5 × 12.5 mm external is 標準).
